Guilfoyle went to the Boston College High School and in 2005, spoke at the commencement of the school’s seniors.

He is best known for his work in CSI: Crime Scene Investigation. Other shows where he has worked in are, Howard the Duck, Crime Story, Miami Vice, Law & Order, New York Undercover, and Ally McBeal. He has around three decades of experience in the movies, with notable works in Three Men and a Baby, Wall Street, Beverly Hills Cop II, Quiz Show, Hoffa, Mrs. Doubtfire, Air Force One, Striptease, Amistad, The Negotiator, Extreme Measures, Primary Colors and L.A. Confidential.

He also made appearances in the HBO movie Live from Baghdad and Alter Bridge's video for their single "Broken Wings".

CSI: Crime Scene Investigation

In CSI, Paul plays the role of LVPD Homicide Detective, Captain James "Jim" Brass, a native of New Jersey, who was earlier posted as the head of the CSI unit in Las Vegas but later sent back to the police homicide division. He struggles with his domestic problems due to his daughter, Ellie who is a prostitute and a drug addict based in Los Angeles. Later though, it is known that he is not the biological father of Ellie. A captain in the homicide division, working with the graveyard shift, he is quite close to the leader, Gil Grissom, and he has even granted a power of attorney to him. He also shares a paternal bonding with Sara Sidle. He is also always at the forefront, conducting interviews of the suspects and examining the witnesses.
